In this situation, we would use 'on' - 'The picture is on the wall'.
The preposition 'in' is used to describe something that is enclosed or surrounded by a specific space such as a box, or a room etc.
The preposition 'on' is used to describe something that is touching the surface of something else. In your example, the back of the picture is touching the wall, which is why we would say, 'on the wall'.
